Analysis of estrogenicity, toxicity and neurotoxicity of Microcystis aerigunosa, Cylidrospermopsis raciborskii and Scenedesmus quadricauda exudates with zebrafish (Danio rerio) embryos.

Popis Cyanobacteria are known to synthesize mixture of diverse biologically active compounds which are partially excreted into natural water bodies. The structural identity as well as potential biological activity and toxicological properties of many of these compounds are still unknown. In this study the zebrafish embryo model was used to investigate the potential estrogenicity, neurotoxicity and general toxicity of exudates from laboratory cultures of two cyanobacteria species Microcystis aerigunosa and Cylidrospermopsis raciborskii and one green algae Scenedesmus quadricauda. Exudate samples were collected after 21-day exponential growth separating the liquid phase from cells; after that the liquid phase was extracted by solid phase extraction. A transgenic zebrafish strain (cyp19a1b-GFP, Brion et al., PloS ONE 2012, e36069) was used to assess the estrogenicity. This strain expresses green fluorescent protein under the control of aromatase B (cyp19a1b) promoter, which is induced by estrogens. The effects of exposure to exudates on locomotor response were used as an indicator of potential neurotoxic effects. A significant increase in estrogenicity compared to control was observed in embryos exposed to 10 times concentrated exudate of Microcystis aerigunosa and 10 and 17 times concentrated exudate of Scenedesmus quadricauda. Phenotypic toxicity was not observed in any concentration of Scenedesmus quadricauda (1 - 17 times concentrated) exudate. However, in case of exudates of Microcystis aerigunosa deformation of tip of the tail appeared from 3.3 times concentrated samples; all embryos in 10 times concentrated exudates had deformities of the tail and frequently also heart edemas and lethality occurred in 17 times concentrated samples. The results of ongoing work on Cylidrospermopsis raciborskii, neurotoxicity and other endpoints will be also presented.
