Popis Many biological compounds exhibit optical activity, which is due to asymmetric carbon on which four different substituents are attached. This configuration leads to two structurally different forms of one substance, when both forms have the ability to rotate the plane of polarized light, one turns to the left side (L-) and the second one to the right one (D-). This phenomena can be monitored by circular dichroism (CD) spectroscopy when the ellipticity is measured in the region of absorption band and this parameter can be used for structural analysis as well as for quantitative determination. However, if L- and D- forms are present in solution in ratio 1:1, i.e. racemic optically inactive mixture, they does not exhibit any rotatation. In our case, the L- and D- forms of tryptophan in aqueous solution were measured. The spectroscopic measurements were done at Jasco J-810 spectropolarimeter (Jasco, Japan) in the wavelength range of 200-300 nm. CD-spectra of pure L- and D-tryptophan and their racemic mixture are presented. It was demonstrated, that CD spectroscopy can be employed for quantitative analysis and determination of purity of chiral aminoacid (Tryptophan).
