Description The spirochete T. pallidum ssp. pallidum is the causative agent of syphilis, a sexually transmitted disease characterized by multiple stages and infection of different organs. It was hypothesized that T. pallidum regulates its gene expression during infection of humans. We have studied the function of sigma-54 factor (RpoN) and sigma-54-dependent promoters. Sigma factors inititate transcription by binding to promoter and to RNA-polymerase. The sigma-54-dependent promoters were identified by the PromScan software. pCS26 plasmid was used for the construction of the transcriptional fusions between the promoters and luxCDABE genes. This reporter system was used for verification of identified promoters. The promoters of polA and tpF1 genes were shown to be dependent on overexpression of treponemal rpoN and fhlA both in wild type E. coli and, more extensively, in E. coli rpoN- or E. coli fhlA-. Regulation of polA and tpF1 genes was confirmed using Q-PCR technique.
