| Description | Overview of the original experimental results: MDMA and methamphetamine developed after repeated administration so called behavioural sensitization to the drug stimulatory effect on locomotion ("open field test"), and antiaggressive influence (agonistic behaviour) in mice. A cross-sensitization to methamphetamine effects was confirmed after pretreatment with morphine and cannabinoid agonist methananadamide. Behavioural sensitization was asscociated with increased relative expression of the genes ("real time PCR") for cannabinoid CB1 and dopamine D1 receptors, but immunofluorescent detetction of CB1 receptors on brain neuronal membranes showed decreased density as susspected phenomenon of internalization. In rat model of I.V. methamphetamine self-administration estrogenized females developed higher sensitization to the drug comparing to males and non-estrogenized female castrates. |
