Popis The aim of this study was to analyze the effect of fish ploidy level and seasonality on innate immune response of tench (Tinca tinca, Cyprinidae), fish species extensively bred in European aquaculture. Fish samples were collected from breeding pond in Vodňany (Czech Republic) during four sampling periods (September 2008, December 2008, March 2009 and June 2009). A total of 81 diploid and 79 triploid specimens of tench were investigated. All hematological parameters measured (erythrocyte count, hematocrit, hemoglobin concentration and leukocyte count) were strongly affected by the season and fish ploidy status. The effect of fish sex was found only in leukocyte count. Blood innate immunity measured by respiratory burst as activity of phagocytes and complement activity were significantly different among different sampling periods (e.g. phagocytes showed lowest activity in winter whereas complement activity was the highest in this period). Moreover, respiratory burst was also influenced by fish ploidy (with lower values in diploids comparing to triploids). Lysozyme activity (higher in autumn and spring) in skin mucus showed significant seasonal changes. However, ploidy had no significant effect on lysozyme. No effect of sex on innate immunity was observed. The level of stress measured by glucose concentration and the concentration of 11-ketotestosterone considered as the steroid hormone affecting immunity only in males showed only seasonal changes and no effect of ploidy was found.
