Popis Background: Recurrent aphthous stomatitis (RAS) is a common chronic inflammatory disease of oral mucosa characterized by single or multiple painful erosions and ulcers. The etiopathogenesis of RAS has not been elucidated yet, but anxiety and stress are considered as one of the main factors influencing the manifestation of RAS. Variability in serotonin transporter gene (SLC6A4), modulating the magnitude and duration of serotonergic neurotransmission, has been previously associated with RAS. The aim of our study was to analyse five polymorphisms in the SLC6A4 gene in patients with RAS and healthy controls. Subjects and methods: Totally, 239 subjects were enrolled in this case-control study: 86 patients with RAS and 153 healthy individuals. The serotonin transporter length polymorphic region (5-HTTLPR) polymorphism, variable number tandem repeat (STin2) and single nucleotide polymorphisms (rs25531, rs1042173, rs3813034) in the SLC6A4 gene were determined by polymerase chain reaction with/without restriction analysis. Results: The allele, genotype and haplotype frequencies of all five studied polymorphisms between RAS patients and healthy controls were similar (P>0.05). Nevertheless, the distribution of allele frequencies of 5-HTTLPR SLC6A4 polymorphism was marginally different between patients with RAS and healthy controls (P=0.07). Conclusions: In contrast to previous studies in Brazilian and Iranian populations, our study did not confirm the association between the SLC6A4 gene variability and susceptibility to RAS.
