Evolution and biogenesis of plant telomerase RNA

Popis The telomerase ribonucleoprotein complex is the most common mechanism for resynthesizing telomeres in eukaryotes. Its central component, the Telomerase RNA (TR) subunit, contrasts with the catalytic Telomerase Reverse Transcriptase (TERT) subunit of telomerase, showing significant divergence in size, sequence, and biogenesis pathways across eukaryotes. Our research uncovered the first bona fide telomerase RNAs in land plants (Fajkus et al. 2019). Contrary to previous assumptions of a polyphyletic origin for plant TRs transcribed by RNA Polymerase II, genuine TRs demonstrate a monophyletic origin not only in land plants but also across the entire Diaphoretickes phylogenetic megagroup (Fajkus et al. 2021). These TRs show conservation of the type-3 RNA Polymerase III promoter which allows to use its Upstream Sequence Element, together with predicted template region of TR, as searchable motifs for TR gene identification. The conservation of TRs' secondary structure enables the efficient use of covariation models to systematically search for TRs across wider phylogenetic ranges (Kilar et al. 2023). Unexpectedly, our recent findings in Hymenoptera and Lepidoptera insects challenged the prevailing concept of a monophyletic origin of TRs in Animalia. Here, we found short TRs of the snRNA type with type-3 RNA Pol III promoters (Fajkus et al. 2023), instead of the RNA Pol II-transcribed H/ACA box sno/scaRNAs, previously considered common for animals. These discoveries illustrate the intricate evolutionary trajectory of TR and suggest a scenario far more complex than previously envisioned.
