Description | Forty eight A. popoffii-like isolates from water sources (e.g. river freshwater, water pond, underground water and stream water) sampled in Czech Republic and Slovakia during 2008-2012, were characterized by PvuII ribotyping (using RiboPrinter identification system), extensive biochemical testing (tube and plate tests, ENTEROtest 24, Biolog system) and MALDI-TOF MS analysis along with reference strains of A. hydrophila group. These strains were tentatively identified as A. popoffii on the base of the negative sucrose acidification, which is a typical trait of this species. MALDI-TOF MS showed the highest discriminatory power for presumptive A. popoffii isolates and this method seems to be valuable for species identification of A. hydrophila group. RiboPrinter demonstrated great heterogeneity of ribotyping patterns among A. popoffii strains but their identification is impracticable due to the absence of A. popoffii reference patterns in the RiboPrinter identification database. Four presumptive A. popoffii isolates (P3282, P4293, P4571 and P4573) displayed very distinct riboprint profiles. Their further investigation using 16S rRNA and cpn60 genes sequencing assigned these strains as Aeromonas veronii and Aeromonas sobria species. |
