Description The increasing prevalence of multidrug-resistant bacteria and the rise of the postantibiotic era show phage therapy as a promising alternative for antibiotic treatment of bacterial infections. The main limitations of the development and production of phage preparations are regulatory requirements, including the quality and effectiveness proof. One of the new strategies to overcome bacterial resistance is the synergy between antibiotics and bacteriophage therapeutics (phage-antibiotic synergy; PAS). The presented work is concerned about new methods of the PAS evaluation. The development of these methods is crucial nowadays, not just because of fulfilling legislative requirements, but also because of the possibility of automatizing the whole process. In this work, the efficacy of two distinct therapeutic phages, 812 and P68, in combination with the three most promising antibiotics in eradication of four Staphylococcus aureus strains, including laboratory and clinical isolates, was evaluated. Experiments were performed under different conditions, e.g. phage:strain input ratios and antibiotic concentrations. All the used antibiotics affect bacterial cell wall metabolism, but their mechanism of action differs. Based on the results obtained by stationary experiments on microbiology dishes, a spectrophotometric method using a TECAN Infinite 200 plate reader followed. The data obtained will be compared with those measured by other methods not previously used for this purpose.
