Description Advancements in preclinical models are pivotal for enhancing our understanding of complex diseases like glioblastoma. Our research team has developed a sophisticated in vitro model utilizing the co-culture of glioblastoma tumoroids and cerebral organoids (termed “GLICO”; Fig.1A), aiming to replicate the tumor microenvironment and investigate glioblastoma cell behavior. Leveraging extensive expertise in stem cell biology and neurodifferentiation, we have differentiated cerebral organoids from healthy induced pluripotent stem cells. We then integrated mature brain organoids with fluorescently labeled glioblastoma cell line U-87 (Fig.1B). This model now facilitates a detailed study of glioblastoma growth dynamics, and migration patterns (Fig.1C), and also allows us to specifically analyze glioblastoma cells after sorting. The pivotal findings, documented in our recent publication [1], underscore the model’s efficacy in mirroring the complex biological processes of glioblastoma, highlighted by significant alterations in glioblastoma cell fate upon interaction with the brain organoids. Efforts are underway to optimize this system for the co-culture of organoids with patients-derived tumoroids to provide a better gateway to discovering potential therapeutic targets and intervention strategies.
