Description | The mechanism of cognitive decline in patients with pharmacoresistant temporal lobe epilepsy has not yet been satisfactorily explained. Even greater uncertainties surround the markedly increased incidence of epileptic seizures in patients with Alzheimer's dementia compared with the age-matched population. Tau protein, amyloid beta, alpha synuclein and TDP 43 are proteins associated with the process of neurodegeneration and their pathological forms and aggregates are pathognomonic of certain types of dementia. A growing body of evidence shows increased abundance of these proteins and their precursors in brain tissue in pharmacoresistant temporal lobe epilepsy. The present project addresses the issue from several perspectives. Electrophysiologically and immunohistochemically, it examines both in vivo and ex vivo brain tissue of temporal lobe epilepsy patients and in vitro organoid cultures of genetically linked Alzheimer's disease. |
