Description | We have developed an azobenzene-based photoactivatable substrate for haloalkane dehalogenases (E.C. 3.8.1.5, HLDs), important enzymes of the ?/ß-hydrolase superfamily. These biocatalysts, which convert halogenated hydrocarbons into alcohols, have wide applications in decontamination, bioremediation, biosensors, pharmaceutical production, and cell and protein imaging. The azobenzene substrate allows detailed analyses of the catalytic mechanism of HLDs and its mutants in a laser-controlled experiment with pre-steady-state kinetics. The new substrate will be used as a detection probe in time-resolved femtosecond crystallography to accurately monitor the structural progress of catalytic reactions. |
